Dopamine Transport – DAT Reviewed at Wikipedia – See the symporter relevance for norepinephrine there as well – genetics matter:

DAT is implicated in a number of dopamine-related disorders, including attention deficit hyperactivity disorder, bipolar disorder, clinical depression, and alcoholism. The gene that encodes the DAT protein is located on human chromosome 5, consists of 15 coding exons, and is roughly 64 kbp long. Evidence for the associations between DAT and dopamine related disorders has come from a type of genetic polymorphism, known as a VNTR, in the DAT gene (DAT1), which influences the amount of protein expressed.2

SERT Transporters

Serotonin Transport – SERT Reviewed at Wikipedia – Genetics and comorbidity abound.

This transport of serotonin by the SERT protein terminates the action of serotonin and recycles it in a sodium-dependent manner. This protein is the target of many antidepressant medications, including those of the SSRI class. It is a member of the sodium:neurotransmitter symporter family. A repeat length polymorphism in the promoter of this gene has been shown to affect the rate of serotonin uptake and may play a role in sudden infant death syndrome, aggressive behavior in Alzheimer disease patients, post-traumatic stress disorder and depression-susceptibility in people experiencing emotional trauma.3

Walsh: On Why MTHFR Genetics Are Limited As Stand Alone Results

Since there are SNPs that tend to reduce methylation and others that tend to increase methylation, a patient’s overall methyl status depends on the overall combined impact of these SNPs. The popular genetic tests for MTHFR, MS, and other SNPs provide interesting information, but are qualitative in nature and limited in their ability to accurately determine overall methyl status. SNPs that reduce SAMe production are often counterbalanced by SNPs that tend toward overmethylation. Diagnosis of overall methyl status is very important in clinical treatment of mental disorders. Two lab assays that directly measure the net effect of the competing SNPs are SAMe/ SAH ratio and whole-blood histamine.4

This diagram will help you understand how supplements and specific amino acids can improve treatment outcomes after testing indicates challenges. With neurotransmitter excess or deficiency, and/or without the balanced dynamic reuptake of neurotransmitters psychiatric medications prove ineffective. Cost effective measurements of these variables often prevent treatment failure.
